Description

Principle of the assay: human Galectin-2 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for Galectin-2 has been precoated onto 96-well plates. Standards(E.coli, M1-E132) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for Galectin-2 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human Galectin-2 amount of sample captured in plate.
Background: Galectin-2 is a protein that in humans is encoded by the LGALS2 gene. It is mapped to 22q13.1. The protein encoded by this gene is a soluble beta-galactoside binding lectin. The encoded protein is found as a homodimer and can bind to lymphotoxin-alpha. It has been found that a single-nucleotide polymorphism in Galectin-2 is significantly associated with susceptibility to myocardial infarction. This genetic substitution affects the transcriptional level of Galectin-2 in vitro, potentially leading to altered secretion of LTA, to which Galectin-2 binds, which would then affect the degree of inflammation
